Discount at LBSs

Website mentions that Cycle Heaven gives members 10% discount on bikes and a couple of others giving discount on accessories with discounts on bikes negotiable at York Cycleworks.

The wife wants her first road bike so is it best to try Cycle Heaven first to take advantage of the discount, or are all shops likely to give around 10% discount if pushed/asked?

We're probably looking at spending around £500-600.

Best thing to do is pay the shops a visit and strike up a conversation with them, kick some tyres, try some bikes and see which models you like/fit.

I wouldn't be led by price first, rather choose a bike that suits your needs/budget and find a shop that's helpful. Price should be the last conversation you have and if you're not happy with the deal walk away and buy elsewhere.

The list of LBSs that do members discount is a good starting point and they each have different models to choose from.

Different member's experiences vary. Of the 2 shops you mention Cycle Heaven are great for the general leisure/commuting cyclist whereas York Cycleworks is perhaps more suited to the club rider/racer, dropped handlebar cycle user....
